Neutrophil Chemotaxis Assay

A robust In vitro assay developed using fresh Neutrophils isolated from healthy donor whole blood. The assay was used to measure compound potency on Neutrophil migration.  The assay utilises Transwell® Permeable Support systems.

Neutrophil Chemotaxis activation

Figure-A: Fold increase in Neutrophil migration over Vehicle response upon addition of FBS (0% to 20%).

Figure-B: Fold increase in Neutrophil migration over Vehicle response upon addition of IL-8, C5a and MCP-1 concentration range (0 to 30 ng/ml).

Neutrophil Chemotaxis inhibition

Here we demonstrate specificity of IL-8 and C5a induced Neutrophil migration using tool compounds. IL-8 strongly induces neutrophil migration that is inhibited by IL-8 antagonist (SCH527123) but not by C5a antagonist (Avacopan). C5a strongly induces neutrophil migration that is inhibited by C5a antagonist (Avacopan) but not by IL-8 antagonist (SCH527123).
